The enzyme uricase (EC1.7.3.3) , or urate oxidase (UO) or factor-independent urate hydroxylase, absent in humans, catalyzes the oxidation of uric acid to 5-hydroxyisourate :
Uric acid + O2 + H2O →5-hydroxyisourate + H2O2 →allantoin + CO2
Uricase is slightly dissolved in alkaline buffer, pH6.3 and the enzyme activity is inhibited by cyanide, Ag+ and Hg2+.
Urate oxidase is mainly localised in the liver, where it forms a large electron-dense paracrystalline core in many peroxisomes. The enzyme exists as a tetramer of identical subunits, each containing a possible type 2 copper-binding site.
Urate oxidase is found in nearly all organisms, from bacteria to mammals , and plays different metabolic roles, depending on its host organism. It was lost in early primate evolution, and so is absent in humans and other higher apes.
The uric acid is hard to be dissolved and the high concentration of uric acid will deposit in the organization to form a stone. When uricase is used for treatment of hyperuricaemia patients, it can reduce the levels of uric acid in the blood, increase allantoate excretion and then eliminate the uric acid strand in the blood and tissues. It is also used to detect the amount of uric acid in the serum and urine.
The uricase provided by our company is recombinant protein. It is of high purity and good activity. The use of recombinant uricase eliminates the risk of virus presence, and other potential adventitious agents found in animal-derived uricase.
Biochemical studies: Uricase can catalyze the enzymatic oxidation of uric acid to allantoate rapidly, so it is used to detect the amount of uric acid in the serum and urine.
