Descriptions

Homocysteine ( Hcy ) is a thiol-containing amino acid formed from methionine during S-adenosylmethionine-dependent transmethylation reactions . It has been demonstrated that even mild or moderately elevated levels of Hcy also increase the risk of atherosclerosis of the coronary , cerebral and peripheral arteries and cardiovascular disease . And currently the hcy level is regarded as the biomarker for cardiovascular disease diagnosis all over the world .

S-adenosyl-L-homocysteine hydrolase (SAHH , EC 3.3.1.1) (AdoHcyase) is an enzyme of the activated methyl cycle, responsible for the reversible hydration of S-adenosyl-L-homocysteine into adenosine and homocysteine.

SAHH is a ubiquitous enzyme which binds and requires NAD+ as a cofactor. AdoHcyase is a highly conserved protein of about 430 to 470 amino acids. The family contains a glycine-rich region in the central part of AdoHcyase; a region thought to be involved in NAD-binding. This protein may use the morpheein model of allosteric regulation.

There are several methods for hcy level detection . And in all these methods the most time-saving , high sensitivity , high throughput is enzyme cycling methods . People has applied this method for hcy diagnosis kit , and this kit is very popular throughout the world .

The principle of the enzyme cycling methods is as the following :

Hcy:  Homocysteine                   SAM:    S-adenosyl-L-methionine HMT: Homocysteine methyltransferase     SAH:  S-adenosyl-L-homocysteine   SAHH: S-adenosyl-L-homocysteine hydrolase  ADA:  Adenosine  Deaminase

The SAHH we can provide is recombinant protein , and it is of high activity and good stability, which can be used for different applications .
