3α-Hydroxysteroid Dehydrogenase (HSD, EC 1.1.1.50) , consisting of 258 amino acid residues with the molecular weight about 26.4 kDa, is a steroid dehydrogenase secreted by Comamonas testosterone.. 3α-hydroxysteroid dehydrogenase can use several kinds of steroid matrix as its substrates, which can reversible catalyze oxidation and reduction reaction of C19-27 steroid 3 hydroxy / ketone radical. The isoelectric point of HSD is 4.8, and its most suitable pH is 7-9. Hg2+, Ag+ can inhibit the activity of the enzyme. Metal ion chelating agent such as EDTA will not inhibit the enzyme activity, On the contrary, it will help the enzyme to keep stability to a certain extent .

Biochemical studies: bile acid is one of the substrates of 3α-hydroxy steroid dehydrogenase. 3α-hydroxy steroid dehydrogenase is used to catalyze the dehydrogenation reaction of hydroxy steroid in clinic. So, HSD is used to detect the total bile acid clinically.
